What You Need to Know

PIK3CA‑Related Overgrowth Spectrum (PROS) is a group of rare conditions caused by a change in a gene called PIK3CA. These changes are not inherited—they happen randomly during early development. Because of this gene change, certain parts of the body may grow larger or differently than others, leading to uneven or extra tissue growth.

Symptoms

Since PROS includes many different conditions, the symptoms can vary. Common symptoms may include: 

  • Uneven or disproportionate growth of at least one body part
  • Vascular malformations
  • Fatty masses or fatty tissue overgrowth
  • Bone and skeletal changes
  • Neurological symptoms including low muscle tone, seizures or developmental differences

Diagnosis

Doctors typically diagnose PROS by a combination of:

  • Clinical evaluation
  • Genetic testing 
  • MRI
  • Ultrasound
